Mrs. Robinson is afkomstig uit de film The Graduate uit 1967. De film gaat over een pas afgestudeerde jongeman die verleid wordt door Mevrouw Robison, een getrouwde vrouw van middelbare leeftijd. Regisseur Mike Nichols wilde graag muziek van Simon & Garfunkel in zijn film gebruiken. Paul Simon zou drie liedjes voor de film schrijven, maar de eerste twee vond Nichols niet goed genoeg. Door het vele toeren met Art Garfunkel was Paul Simon niet aan een derde nummer toegekomen. Uiteindelijk speelde hij een liedje voor aan Nichols dat Mrs. Roosevelt heette, naar Eleanor Roosevelt, de echtgenote van Franklin D. Roosevelt. Nichols zei dat Paul Simon de titel moest veranderen in Mrs. Robinson en gebruikte het vervolgens in zijn film, naast oudere nummers van het duo, zoals The Sound Of Silence en Scarborough Fair. De versie van Mrs. Robinson in de film en op de soundtrack verschilt van de hitversie. In de film wordt alleen het refrein gezongen en de tekst daarvan is iets anders. Pas een paar maanden na de film werd de definitieve versie van Mrs. Robison op single uitgebracht. Daarin is nog te horen aan regels als "Going to the candidates' debate" dat Paul Simon bij het schrijven presidentsvrouw Eleanor Roosevelt in zijn hoofd had.

Wist je dat? In het laatste refrein wordt in plaats van Mrs. Robinson Joe DiMaggio genoemd. Joe Dimaggio (bijgenaamd Joltin' Joe) was een Amerikaanse honkballer die van 1936 tot 1951 bij de New York Yankees speelde. Joe DiMaggio was aanvankelijk niet blij dat hij in het nummer werd genoemd. Hoewel de tekst zegt: "Where have you gone, Joe DiMaggio" en "Joltin' Joe has left and gone away", vond DiMaggio zelf dat hij nooit weggeweest was. Paul Simon wist hem er in een persoonlijk gesprek echter ervan te overtuigen dat deze regels bedoeld waren als een eerbetoon. Paul Simon was eigenlijk fan van de honkballer Mickey Mantle, maar gebruikte de naam Joe Dimaggio, omdat die beter in het metrum paste. In 1969 scoorden Booker T. & the MG's een bescheiden Top 40-hit met een instrumentale uitvoering van Mrs. Robinson.
